胡文浩现任中山大学教授、药学院院长。国家杰出青年基金获得者、教育部“长江学者”特聘教授。先后在四川大学获得学士学位(1987)、中科院成都有机所获得硕士学位(1990)和香港理工大学获得博士学位(1998,导师:陈新滋院士),随后在美国亚利桑那大学从事博士后研究(1998-2001)。胡文浩教授先后在美国Genesoft 和Bristol-Myers Squibb 公司担任高级研究员,于2006年回国任华东师范大学化学系教授、上海市高校“东方学者”特聘教授(2009)和上海分子治疗与新药创制工程技术研究中心主任(2011-2016),2016年8月调入中山大学任药学院院长,广东省“珠江人才计划”创新创业团队带头人,广东省手性分子与药物发现重点实验室主任(2019起)。研究方向主要集中在不对称催化多组分反应、手性药物发现和药物合成工艺,至今已在Nature Chemistry, Nature Communications, Science Advances, Journal of the American Chemical Society, Angewandte Chemie International Edition和Journal of Medicinal Chemistry等期刊上发表论文300余篇,授权发明专利50余项。
Dr. Wenhao Hu is currently a professor and dean of the School of Pharmacy at Sun Yat sen University. He received National Natural Science Funds for Distinguished Young Scholar (2011) and was selected as The Chang Jiang Scholar of Ministry of Education of China (2014). Hu received his B.S. in chemistry from Sichuan University (1987), and Ph.D. from Hong Kong Polytechnic University with Professor Albert S. C. Chan (1998), and became a postdoctoral fellow at University of Arizona with Professor Michael P. Doyle. During 2002-2006, he worked as a medicinal chemist at Genesoft in South San Francisco and a process chemist at Bristol-Myers Squibb in New Jersey. In 2006, Prof. Hu joined East China Normal University as a Distinguished Professor of Shanghai Oriental Scholar (2009-2016) and a director of Shanghai Engineering Research Center of Molecular Therapeutics and New Drug Development (2011-2016). In August 2016, he moved to Sun Yat-sen University as the dean of School of Pharmaceutical Sciences via Innovation and Entrepreneurship Team of “The Pearl River Talent Recruitment Program" of Guangdong Province. Prof. Hu’s research interest focuses on novel multi-component reactions, medicinal chemistry, and process chemistry. He has published over 300 research papers in international journals, including Nature Chemistry, Nature Communications, Science Advances, Journal of the American Chemical Society, Angewandte Chemie International Edition and Journal of Medicinal Chemistry, etc.
讲座摘要/Abstract:
多组分反应因其操作简便、合成效率高、原子和步骤经济性等优点,在现代有机化学和药物发现研究中发挥了重要作用。多组分反应的组分多,其反应途径复杂,选择性控制困难,特别是对映选择性控制极具挑战。胡文浩课题组基于利用第三种组分捕获由两种组分生成的活性中间体(叶立德或者两性离子对),改变传统的两组分反应途径,探索多组分反应的选择性调控规律,发展一系列新型实用的不对称催化多组分反应[1]。发展多种催化剂协同催化体系,调控反应的选择性,实现了一系列手性多官能团化合物的高效合成;通过对活泼中间体反应活性的研究,理解反应机理,丰富活性中间体理论;通过中间体捕捉中间体的策略,发展高阶多组分反应策略[2,3];应用多组分反应完成了高效合成紫杉醇衍生物,极大地提高了合成效率;通过与人工智能交叉融合,拓展可及化学空间,构建合成可及化合物库,加速苗头/先导化合物的发现。
By virtue of exceptional competence for the rapid construction of polyfunctionalized scaffolds in one-pot operation, multicomponent reactions represent one of the most efficient and practical synthetic methods.[1] One of the research interests in Hu group at Sun Yat-sen University is developing novel and efficient multicomponent reactions via carbene gem-difunctionalization and applying these reactions to the synthesis of natural products and pharmaceuticals. In this talk, an efficient and highly enantioselective four-component reaction via convergent assembly of two in-situ generated intermediates will be present.[2,3] The highly diastereoselectivity and enantioselectivity of the novel multicomponent reaction are achieved by cooperative catalysis. Application of the multicomponent reaction in total synthesis of taxol and its derivatives will also be presented.
