Conformational dynamics drives integrin avb8 mediate L-TGF-b activation
讲座摘要/Abstract:
In the era of single particle cryo-EM and AI driven protein structure prediction, determining high-resolution structure is no longer a difficult but an achievable goal. However, study and understand protein dynamics is still a challenging task. Using avb8 mediated L-TGF-b1 activation as an example, I intend to illustrate the significance and challenging of studying protein dynamics, when conformational dynamic is part of protein function. TGF-b, essential for development and immunity, is expressed as a latent complex (L-TGF-b) non-covalently associated with its prodomain and presented on immune cell surfaces by covalent association with GARP. Binding to integrin avb8 activates L-TGF-b1/GARP. The dogma is that mature TGF-b must physically dissociate from L-TGF-b1 for signaling to occur. Our previous studies discovered that avb8-mediated TGF-b autocrine signaling can occur without TGF-b1 release from its latent form. We reveal a dynamic allosteric mechanism of autocrine TGF-b1 signaling without release where avb8 binding redistributes the intrinsic flexibility of L-TGF-b1 to expose TGF-b1 to its receptors.
