Rui Fang is a graduate student in Ying Lu lab at the department of Systems Biology, Harvard Medical School, where she is investigating how the ubiquitin-proteasome system helps to dismantle the intracellular protein aggregates by integrating biophysical, biochemical and cell biology techniques with mathematical modeling. She is broadly interested in developing therapeutic approaches to combat pathogenic protein aggregates in aging and neurodegeneration. She received her BS in the Integrated Science Program from Peking University in 2018 and will receive her PhD from Harvard University in 2023.


讲座摘要/Abstract:

Proteostasis is maintained by the ubiquitin-proteasome system targeting aberrant proteins whose accumulation and aggregation are hallmarks of aging and neurodegenerative disorders. However, the mechanism of how protein aggregates are processed, as well as which factors are involved, are mostly elusive. In this talk, I will present two studies investigating the mechanisms of the 26S proteasome-mediated protein degradation and the "aggresome", a macroscopic aggregate deposition site. We developed a novel approach to simulate the proteasome's conformational dynamics using its empirical free-energy landscape (FEL). The FEL model successfully predicted the proteasome behaviors under various conditions and provided mechanistic insights. I also established the first in vitro model system to recapitulate the multiple stages of the aggresome pathway including nucleation, transportation, and clearance, which provides unprecedented opportunities to identify the key players and their mechanistic activities in each stage. Together, my work will help reveal how different components in the protein quality control systems collaborate to mount an effective defense against protein aggregation and inspire novel therapeutics.
