Fabrication and Measurement of Human Organ-on-a-Chip Systems
讲座摘要/Abstract:
Human organ-on-a-chip systems (also known as microphysiological systems, MPS) are innovative platforms that integrate technologies such as stem cells, organoids, tissue engineering, and microfluidics to simulate human tissue and organ functions as well as physiological environments on microchips. These systems hold significant promise for fundamental disease modeling, drug screening, and precision medicine. Around 2012, both the United States and China initiated organ-on-a-chip research programs respectively, accelerating scientific progress and industrial translation in this field. In 2016, organ-on-a-chip technology was recognized by the World Economic Forum as one of the top ten emerging technologies. In 2022, the world’s first new drug developed entirely based on organ-on-a-chip data progressed into clinical trials, marking a milestone that promoted several countries to begin formulating relevant legislation. In China, the first organ-on-a-chip space experiment was completed in 2023. In 2024, the National Medical Products Administration (NMPA) released technical guidelines for the application of organ-on-a-chip technology in rare disease drug development, and in the same year, the national standard for skin-on-a-chip systems was officially issued and implemented. This report will focus on recent advances made by our team in the areas of chip fabricated two-photon polymerization, conductive polymer-based extracellular matrix (ECM) materials, photonic crystal cellular force microscopy, and modular “liquid LEGO” chip design and fabrication. Furthermore, we will present the applications of organ-on-a-chip platforms in scientific research and biomedicine.
